Is the national insurance card required?

Does not have a national insurance card, will it affect naturalization? However, it is optional in the online application form.

An NI card is not required - do they exist?
If you got an NI number then just quote it on the form - otherwise not an issue as it's an optional field.

biometric information

Can I continue to use the biometric information that was applied when I applied for permanent residence 2 years ago?

You have to enroll biomterics information again at UKCVAS when you apply for citizenship.

It can be anyone and any nationality age 25 or over.

Core Service Point
Enhanced Service Point
Premium Lounge Service Point
Their main difference? What kind of appointment is easy?

For naturalisation you basically just have one appointment, to give your biometrics. They are not interviews.
You can purchase extra services such as upload of documents, though most applicants do it themselves for free.
Refer to the UKVCAS website for more details. Note UKVCAS are a contractor to Home Office UKVI and not caseworkers - they do not review or approve your application. Note also that for naturalisation there is no expedited or same day service.

Your spouse may or may not be a dependent from an immigration point of view, i.e. you've ILR and she/he's a dependent drawing her immigration status from yours or you've IRL and she/he's ILR or another immigration status of their own not drawn from yours, and is certainly not a dependent by definition as most children would be, minors anyway, both by definition and in as far as immigration law is concerned. If your spouse is not applying with you would be obvious from your application anyway, if you have minors not applying with you wouldn't unless this specific question is asked.

Presumably you've included your wife's details where it asks you to provide these? And, you've no children? If so, N would be the correct answer in my view.
