I have applied for eea2.just i want to know that Is EEA family member held comprehensive sickness insurance required for rc?

Yes it is required if EEA national is not a worker or self employed.

ravi thanx for reply.she is working in company.so it means she dont need comprehensive sickness insurance.
one thing more i want to ask i send her NHS card and my NHS card with the application.it does any negative affect on my application

Your wife is a worker so you don't need CSI
As you attached NHS card,if both NHS cards are on same address then it may be consider as a proof of address.In some cases NHS cards are acceptable as a proof address.
