Hi,
I am going to apply for my wife british citizenship. I am using my employer as a reference.
In question 5.2 Say how you know the applicant, he is writing Employee's wife.
Although, my employer did not met my wife before. Is it OK?

you have to use a referee that has known your wife personally for at least 3 years, so no, your employer wont be acceptable.

You can't apply for your wife. She needs to apply herself.

But my boss does know about my wife for last 3 years.

I have lots of friends and colleagues who know I have a husband, but they don't know him personally. It does not say 'referees should know ABOUT the applicant' nor does it say 'referee should know ABOUT applicant because referee knows applicants spouse'
You cannot use a referee that has never met the applicant. Referees are contacted at random if necessary.
5.1 One referee should be a person of any nationality who has professional standing, eg
minister of religion, civil servant, or a member of a professional body e.g. accountant or
solicitor (who is not representing you with this application). The other referee must normally be
the holder of a British citizen passport and either a professional person or over the age of 25.
Both should declare that:-
• they are not a relative, solicitor or agent of the applicant, or related to the other referee;
• they are not employed by the Home Office;
• they have not been convicted of an imprisonable offence (unless that conviction can be disregarded in line
with the table shown on page 16 of the Booklet AN);
• they have known the applicant personally for more than 3 years;
• they are willing to give full details of their knowledge of the applicant;
• they will advise the Home Office of any reason why the applicant should not be naturalised.

My wife met him multiple times on christmas party each year.
So my question is that what shall i write in How do you know the applicant

My answers are based on your statements above. You need to clarify and explain the details correctly otherwise it just wastes time and causes misunderstanding.
Are you British?? How long has your wife been resident in the UK??

My wife did met him every year on christmas party.
I am british national. She has been resident in UK for last 3 years.
My employer had already said yes to become referee for my wife application

And does your wife not have any other professional she can use as a referee that has known her
You have stated employer has not met her but does know about her and now you are saying employer has met her 1 a year over 3 years.
I would be suspicious if I was a case worker to be honest, but it is her application (not yours) and if she is happy with your employer being her referee then that is her business.
