My friend is going to apply for Naturalisation next week. he had a letter on the over speeding car but no penalty or points. They asked him to attend training. Whether he needs to disclose this matter or not?

As long no fine or points were given by the court, your friend is fine.
Declare the speed awareness course but it will have no impact on the AN application.
