28 day rule and priority service?

The 3-year qualifying period for my exceptional talent visa gives a 4 October 2021 date. 28 days before that would be 7 September 2021. Can I apply for ILR on 7 September and get this done with priority service? I'm confused because priority service would be 5 days which means the visa will be processed by 12 September (or around that date..well before I actually complete the 3 years qualifying period).

Yes, you can. There is no such a thing as 28 days rule. Home office procedures allow you to apply within 28 days and ILR is granted as a concession if you are eligible.
