If someone is on tier2 and 5 years are completed on 24 October then are they allowed to launch and submit application after 27 september which is 28 days before the date?

Is it the date when your online application is submitted or when the in person appointment takes place?

The 28 day rule applies for both postal and PEO appointments.

so i can book before 27 september which is even more than 28 days but my appointment must be after 27th september so that actual appointment is at max only 28 days before the total 5 year period completes.

Yes, it is not the date that you book the appointment.
