Work Permit - where should i apply frm?

some background :
Indian citizen;have a job offer in software development ; have obtained a LMO-Labour Market Opinion exemption letter ( thus - don't need an HRSDC approval) - and am applying under facilitated processing for IT workers; have visitor status in Canada currently.

question/s:
1. should i apply from a consulate in the US or from New Delhi (India); i dont have a visa to enter the US...SO can't go there for an interview if reqd. But mailing is an option.
2. will the fact that I've already applied for a PR frm Buffalo affect granting of work permit...from a consulate in the US / India?

would appreciate any other helpful pointers....
