Kindly advise, what should be consider as application date?

Is this when you pay it online or biometric appointment at Sophera?

My visa is due to expire on 27-04-2019 so do I have book my appointment before 27th with Sophera ??

Yes. UKVCAS appointment date is irrelevant. Your application date is the date of online submission.

Also, as mentioned earlier visa expiry is 27th so can I book my appointment go to UKVCAS first week of May?

Date of application is the date you submit and pay for the online application form.

You get 45 working days to book an appointment. You can choose the date and time you wish depending on availability and the centre you choose. You can get free appointments at a core centre. You have to pay for evening/weekend/next day appointment.
If you apply around 26th, highly likely you can secure a free appointment by 1st week of May at a core centre.
