Moving from TIER-5 Govt Authorised Programme to TIER-2

I am currently working on TIER 5 Govt. Authorised Programme and recently get offer on TIER-2. My employer HR department offered me TIER-2 Sponsorship to apply with UKBA from inside UK.

My question is do i need to apply for TIER-2 from home country or i can apply while staying inside UK.
