I applied for entry clearance in 2008 under Tier 1 General and proved knowledge of English Language with a letter from my University back home confirming that my degree was taught in English. The letter was however retained at the Home Office then.

I'm applying for settlement in a few weeks time and I'm confused with this new law about KoLL. Do I need to get another letter from my home University confirming that my degree was taught in English (again). Or do I just assume that since I had earlier shown evidence I don't need to do that again?

You will not need it again, but may need to draw to their attention that you provided that to the ECO as evidence that you meet the requirement. If you have the letter again, no harm in resending for settlement.
