Hi, I am in UK from past 1 year as a Tier 2 ICT long term dependent, Is it possible to convert my visa to a Tier 2 general visa if a company is ready to offer , is there any cooling period for converting

No, there is no cooling off period for PBS dep changing to Tier 2 G.

You have to apply from your home country for entry clearance. You cannot 'switch' or 'convert' within the UK.

Your sponsoring employer must complete the full and proper RLMT process before they can issue a CoS
