No, because when your wife switched to a FLR(M) visa her immigration clock was set to zero, putting her on a new 5 years route to settlement.
She will have to apply for another FLR(M) extension of 2.5 years before qualifying for ILR. In addition to the visa fee, she will also have to pay the £500 NHS surcharge.
