I got few doubts regarding settled status please I’m hoping someone from here can help me.
I got married to EU citizen on 2014 October. And my 1st application got rejected due to home office can’t find her work place and I got RC on second time that’s on 2018 February so when I’m eligible for applying my settled status? And same time can I apply for permanent residence application as well please?

this previous thread, so not going to answer this again. Kindly refrain from asking the same things twice, especially when people have already spent time answering your questions in detail. If something is not clear, then next time kindly ask questions in your original thread, no need for duplication.
Recently I have lost my RC card while travelling and I reported to police and home office. So do I need to re apply for my RC again or wait to apply to apply for permanent residence card directly?
You can choose not to replace your RC and to wait until you apply for permanent residence, but you will likely find yourself locked inside the UK without being able to travel abroad or prove your right to work in the UK, as you will not have any document proving these rights. You will not be illegal though, as long as you continue to be married and your spouse continues to exercise their treaty rights, or if they already have permanent residence. So if you are not planning to change jobs, move houses, or leave the country in between, you could just wait (although you never know what life throws at you, not having documentation it's a risk you take).
